Inhoud

The course provides the students with insights in how the three-dimensional (3D) structures of macromolecules can be determined by the three main methods in Structural Biology: Nuclear Magnetic Resonance (NMR), X-ray crystallography and Electron Microscopy (EM). The merits and shortcomings of each of these analytical methods are discussed in detail, as well as how they can be combined to provide an accurate picture of macromolecular conformation, dynamics and interactions. Hybrid approaches where data from other methods such as Mass Spectrometry are included are also briefly covered. The computational aspect of Structural Biology, where the experimental data is transformed to an in silico model of the macromolecule, is a core part of this course, as is validation of the quality of the obtained 3D models.

Participation to the practicals is obligatory. For each unjustified absence, two points will be subtracted from the mark on the practicals. Unjustified absence for more than half of the practical sessions leads to exclusion from participation to the oral exam. No repetitions can be organized for the missed sessions, and the subtracted points are therefore always transferred to the August/September exam session.

General Competencies

The student understands the basis of NMR, X-ray crystallography and Electron Microscopy.

The student is knowledgeable about which analytical techniques to apply for which problems in structural biology, and understands how to execute this practically.

The student knows the relationship between an atomic model and Structural Biology data.

The student has theoretical and knowledge on how to calculate a 3D structure of a macromolecule and has gained practical knowledge for structure determination using X-ray crystallography and cryo-EM and how this process is validated.

The student can read an article on the structure determination by a variety of methods in Structure Biology, understand and assess the quality of the structure described herein.

The student knows how the dynamics of a protein can be studied and its usefulness in biological processes.

The student learns various applications of Structural Biology, such as determination of protein interactions.

Beoordelingsinformatie

De beoordeling bestaat uit volgende opdrachtcategorieën:
Examen Mondeling bepaalt 80% van het eindcijfer

Examen Schriftelijk bepaalt 20% van het eindcijfer

Binnen de categorie Examen Mondeling dient men volgende opdrachten af te werken:

  • Oral examination with written met een wegingsfactor 1 en aldus 80% van het totale eindcijfer.

  • Report evaluation met een wegingsfactor 1 en aldus 20% van het totale eindcijfer.

Evaluation by a written report during the course and an oral examination with written preparation.

The mark for the exam is composed of two parts: one mark for the oral exam with written preparation (weight 80%) and one mark for the practicals (under the form of an individual report - weight 20%). Students need to pass both on the oral exam and on the report for the practical sessions. If a student fails on at least one of these, the lowest mark is taken as the final mark.

The oral exam consists to three open questions, one from each part of the course and connected to one of the instructors. All tree questions have equal weight. In order to pass, one needs to pass for at least two questions, and no questions should lead to less than 6/20. If these conditions are not fullfilled, the final mark for the oral part of the exam corresponds to the mark of the question that was scored lowest.

Students that fail in the January exam session can ask to be exempt from the report or the oral exam if he/she passed this part. When the student failed on the report, a new report will be asked, but any penalization due to unjustified absence will be retained. The report deals with the individual practical work that was carried out during the WPO sessions and therefore cannot be complete if WPO sessions were missed.

A student who does not pass in both the January and August/September sessions will have to repeat the whole course including both practicals + report and oral exams.
